    摘要:

    目的观察七氟烷吸入麻醉后老年糖尿病大鼠的认知功能变化,并探索其神经递质谷氨酸(Glu)/γ-氨基丁酸(GABA)的变化规律。方法清洁级健康SD老年大鼠63只,高糖高脂饲料复合链脲佐菌素腹腔注射建立糖尿病模型,Morris水迷宫实验选取学习记忆能力下降的大鼠,观察吸入3%七氟烷1,2,3 h后记忆能力的变化和海马组织中Glu和GABA的含量改变。结果建立糖尿病模型后,老年大鼠学习、记忆功能均下降,海马Glu降低,GABA升高,Glu/GABA比值降低(P<0.05);吸入七氟烷后学习记忆能力、Glu/GABA比值降低更为明显(P<0.05﹚,吸入3 h后的差别更显著。结论长时间吸入高浓度(3%)七氟烷可进一步降低糖尿病相关认知功能降低的老年大鼠的记忆能力,其机制可能与海马中神经递质Glu和GABA失衡有关。

    Abstract:

    ObjectiveTo study the effect of sevoflurane on cognitive function in the diabetes-associated cognitive decline(DACD)aged rats and explore its mechanism in role of Glu and GABA in hippocampus.MethodsSixty three SD rats, aged 20 months and weighting 300~400 g, were fed by the high sugar and high fat for 16 weeks and composite intraperitoneal injection of streptozotocin (STZ) to establish model of DACD, followed by the 3% sevoflurane inhalation 1, 2 or 3 h.5 days later the level of cognitive function were measured again by Morris water mazeand and the content of Glu and GABA in hippocampal were determined using HPLC.ResultsFed by high sugar high fat composite injected STZ could successfully established DACD model.The level of cognitive function in Group sevoflurane inhalation were statistically reduced (P<0.05), the content of Glu were reduced and GABA were increased and the ratio of Glu/GABA were reduced (P<0.05, respectively).Conclusion3% sevoflurane inhalated more than 3 hours induced short term cognitive impairment in the DACD aged rats, the mechanisms might associate with the out-of-balance of Glu and GABA in hippocampus.
