    摘要: 目的 建立小鼠睾丸注射模型,获得一种蛋白抑制的简便易行、高效且存活率高的方法,为小鼠精子发生的相关实验研究提供借鉴。 方法 通过腹腔手术暴露小鼠睾丸,睾丸内进针注射蛋白抑制剂,间断缝合法缝合切口。 结果 组织学结果观察发现,实验抑制组小鼠睾丸生精小管内精母细胞染色体排列紊乱,存在中期阻滞现象,无法正常分裂。此模型实现了小鼠睾丸内蛋白的功能抑制,且小鼠存活率高。 结论 小鼠睾丸抑制方法常用阴囊注射法,手术时间局限性大。小鼠腹腔手术睾丸注射法可实现3周龄小鼠睾丸内蛋白抑制,实验成功率高,抑制效果好,有利于精母细胞减数分裂相关研究。

    Abstract:

    AABSTRACT: Objective A mouse testicular injection model was established to obtain a simple, efficient and high survival rate method of protein inhibition, which can be used as a reference for related experiments and studies of spermatogenesis in mice. Methods The testes of mice were exposed by abdominal operation. Protein inhibitors were injected into mouse testes. The wound was sutured intermittently. Results Histological results showed that the chromosomes of spermatocytes in seminiferous tubules of the inhibition group were misaligned. The spermatocytes were arrested at metaphase and could not divide normally. This model can inhibit the proteins in the testes of mice, and the survival rates of mice were high. Discussions The commonly used method of testicular inhibition in mice is direct injection through the scrotum, however, the time window for operation is limited. Testicular injection through abdominal surgery in mice can achieve testicular inhibition in the 3-week-old mice with high success rate and good effects on protein inhibition. This method is suitable for the studies of male meiosis in spermatocytes. KEY WORDS: abdominal surgery; mouse testis; inhibition model; spermatogenesis
